Doxycycline hyclate
Indications/Uses
Treatment of variety of infections eg, Rickettsial infections including ehrlichiosis, typhus, spotted fever, trench fever & Q fever; chlamydial infections including psittacosis, lymphogranuloma venereum, trachoma, non-gonococcal urethritis; mycoplasmal infections especially pneumonia; Legionnaires' disease; gonorrhea & syphilis; surgical infections prophylaxis. Prophylaxis of leptospirosis.
Dosage/Direction for Use
Adult 200 mg as single dose on 1st day or 100 mg repeated after 12 hr followed by 100 mg daily. Childn >8 yr weighing ≤45 kg Initially 4 mg/kg in 2 divided doses followed by 2 mg/kg daily. Patient w/ sensitive gonococcal infections 300 mg as single dose or followed by a 2nd similar dose 1 hr later. Patient w/ syphilis 200 or 300 mg daily for at least 14 or 10 days respectively. Acne 50 mg daily. Relapsing fever & louse-borne typhus 100 or 200 mg as a single dose. Prophylaxis of scrub typhus 200 mg as a single dose. Prophylaxis of leptospirosis 200 mg once every 7 days. Should be initiated 1-2 days before & throughout exposure.
Administration
May be taken with or without food: Take w/ at least 1/2 glass of water.
Special Precautions
Oesophageal ulceration if taken w/ insufficient fluid or in recumbent posture. May cause permanent teeth discoloration in childn. Pregnancy & lactation.
Adverse Reactions
Nausea, vomiting, diarrhea, dry mouth, glossitis, tongue discoloration, stomatitis, dysphagia, oesophageal ulceration, oral candidiasis, vulvovaginitis, pruritus, enterocolitis, severe uraemia & hepatotoxicity.
Drug Interactions
Reduced absorption w/ divalent & trivalent cations eg, Al, bismuth, Ca, Fe, Mg & Zn; concomitant administration w/ (antacids, Fe prep, milk & dairy products); Na bicarbonate, colestipol & kaolin-pectin. Nephrotoxicity may be exacerbated by diuretics, methoxyflurane & other nephrotoxic drugs.
