Gabapentin

Indications/Uses

Adjunctive therapy for partial seizure & partial seizure w/ secondary generalization in patients who are not controlled w/ standard anticonvulsant as monotherapy or combined therapy, or in patients who are intolerant to therapeutic doses of these drugs; simple & complex partial seizure, especially secondary generalized tonic-clonic seizure.

Dosage/Direction for Use

Adult & childn >12 yr Effective dose: 900-1,800 mg daily. Dose titration: Day 1: 300 mg once daily. Day 2: 300 mg twice daily. Day 3: 300 mg 3 times daily. Dose may be increased to 1,200 mg daily in 3 divided doses, & may continue titration using increments of 300 mg daily in 3 divided doses until max of 2,400 mg daily. Max interval of 3 times daily dose: 12 hr. Impaired kidney function CrCl >60 mL/min 400 mg 3 times daily. Total daily dose: 1,200 mg, 30-60 mL/min 300 mg twice daily. Total daily dose: 600 mg, 15-30 mL/min 300 mg once daily. Total daily dose: 300 mg, <15 mL/min 300 mg daily. Total daily dose: 150 mg. Patient undergoing hemodialysis 300-400 mg loading dose. Maintenance dose: 200-300 mg every 4 hr of hemodialysis.

May be taken with or without food.

Hypersensitivity.

Special Precautions

Not considered effective in treatment of seizures & may aggravate seizures in some patients. May trigger status epilepticus in abrupt w/drawal. Patients who experienced mixed seizure, including absence seizures. Concomitant use w/ other antiepileptic medications.

Adverse Reactions

Weakness, pain, fatigue, wt loss, facial edema, chest pain; flatulence; purpura; vertigo, hyperkinesia, decreased or loss of reflex; agitation; bronchitis, sinusitis, pneumonia; laceration, maculopapular rash; UTI; visual changes, common visual disturbances; increased plasma conc of antiepileptic drugs; drowsiness, ataxia, dizziness, fatigue, nausea &/or vomiting.
