Co-amoxiclav (amoxicillin & clavulanic acid). Per 5 mL Amoxicillin trihydrate 400 mg, clavulanate K 57 mg
Indications/Uses
Short-term treatment of common bacterial infections eg, URTI including ENT (eg, tonsillitis, sinusitis, otitis media), lower resp tract (eg, acute & chronic bronchitis, lobar & bronchopneumonia), GUT (eg, cystitis, urethritis, pyelonephritis, female genital infections), skin & soft tissue, bone & joint (eg, osteomyelitis), & other (eg, septic abortion, puerperal & intra-abdominal sepsis, septicaemia, peritonitis & post-surgical infections) infections. Prophylaxis against infections which may be associated w/ major surgical procedures eg, GI, pelvic, head & neck, cardiac, renal, joint replacement & biliary tract.
Dosage/Direction for Use
Childn >6 yr (18-40 kg) 5 mL tid, may be increased up to 50 mg/kg daily in divided doses every 8 hr in severe infection, <12 mth 25 mg/kg daily eg, 7.5-kg child would require 2 mL tid.
Administration
May be taken with or without food: Add 64 mL of water & shake. Best taken at start of meal for better absorption.
Contraindications
History of hypersensitivity to β-lactams eg, penicillins or cephalosporins. Patients w/ previous history of co-amoxiclav associated jaundice/hepatic dysfunction.
Special Precautions
Not to be used w/ drugs that delay peristalsis eg, opiates & diphenoxylate w/ atropine. Provide fluids, electrolytes & protein replacement therapy when indicated. Hepatic dysfunction. Contains aspartame; patients w/ phenylketonuria. Avoid use during pregnancy especially 1st trimester. Lactation.
Adverse Reactions
Gastritis, stomatitis, glossitis, black "hair" tongue, indigestions, nausea, vomiting, diarrhea, enterocolitis, pseudomembranous colitis & candidiasis; skin rashes & urticaria; transient hepatitis & cholestatic jaundice; anemia, thrombocytopenia, thrombocytopenic purpura, eosinophilia, leukopenia & agranulocytosis; phlebitis at inj site.
Drug Interactions
Decrease in plasma conc of total conjugated estriol & estradiol. Increased incidence of rashes w/ allopurinol. May precipitate disulfiram-like (antabuse) reaction w/ alcohol. Amoxicillin: Decreased renal tubular secretion that may result in increased & prolonged blood levels w/ probenecid.
