Description
Neuro-B's: Each capsule contains: Thiamine Mononitrate (Vitamin B1) 300 mg, Pyridoxine Hydrochloride (Vitamin B6) 100 mg, Cyanocobalamin (Vitamin B12) 100 mcg.
Neuro-B's 500: Each film-coated tablet contains: Thiamine Mononitrate (Vitamin B1) 500 mg, Pyridoxine Hydrochloride (Vitamin B6) 250 mg, Cyanocobalamin (Vitamin B12) 1 mg.
Action
Neuro-B's: Vitamin B replacement: Vitamin B1 and B6 constitute part of an enzyme system in the nerve cell and are necessary for normal function of nerve tissue. They are used in the treatment of neurological disorders associated with nerve cell dysfunction. Vitamin B12 is necessary for the synthesis of nucleic acid in the erythropoiesis and plays an important role in the metabolism of the nerve tissue.
Neuro-B's 500: Pharmacology: Pharmacokinetics: Thiamine Mononitrate (Vitamin B1) in small amounts are well absorbed from the gastrointestinal tract but absorption of dose larger than about 5 mg is limited. It is widely distributed to most body tissues, and appears in breast milk. Thiamine is not stored to any appreciable extent in the body and amounts in excess of the body's requirements are excreted in the urine as unchanged thiamine or as metabolites.
Pyridoxine Hydrochloride (Vitamin B6) is readily absorbed from the gastrointestinal tract and is converted to the active forms pyridoxal phosphate and pyridoxamine phosphate which are mainly stored in the liver. These are oxidized into 4-pyridoxic acid and other inactive metabolites which are excreted in the urine.
Cyanocobalamin (Vitamin B12) binds to intrinsic factor and is then actively absorbed from the gastrointestinal tract. Absorption is impaired in the absence of the intrinsic factor, with a malabsorption syndrome, with the disease or abnormality in the gut, or after gastrectomy. Cyanobalamin is stored in the liver, excreted in the bile, and undergoes extensive enterohepatic recycling; part of an administered dose is excreted in the urine, most of it in the first 8 hours.
Indications/Uses
Neuro-B's: Painful neurological manifestation e.g. neuritis, polyneuritis, neuralgias, lumbago, ischialgia, sciatica, cervical and shoulder-arm syndrome, rheumatic pains. Herpes zoster, encephalopathies. Neuropathies caused by certain diseases states e.g. diabetes and cardiac disorders. Alcoholic neuropathy; Iatrogenic complications arising from INH, reserpine and phenothiazine therapy; other drug induced neuropathies. Hyperemesis gravidarum; neuropathic changes during pregnancy.
Neuro-B's 500: For the treatment and prevention of Vitamin B1, B6, and B12 deficiency.
Dosage/Direction for Use
Neuro-B's: For therapeutic use: 2 to 4 capsules daily until acute symptoms subside. Chronic cases may require longer therapy depending on the needs of the patient. For prophylactic administration when disease or drugs are likely to lead neurological complications: 1-2 capsules daily. Or as prescribed by the physician.
Neuro-B's 500: Adults: 1 tablet daily or as prescribed by a physician.
Special Precautions
Neuro-B's 500: Thiamine Mononitrate (Vitamin B1): Anaphylactic reaction can occur with IV use.
Pyridoxine Hydrochloride (Vitamin B6): long term high dose administration of pyridoxine has been associated with the development of severe peripheral neuropathies.
Cyanocobalamin (Vitamin B12) should, if possible, not be given to patients with suspected Vitamin B12 deficiency without first confirming the diagnosis and should not be used to treat megaloblastic anemia of pregnancy. Regular blood monitoring is advisable. Administration of Vitamin B12 doses greater than 10 mcg daily may produce a hematological response in the patients with folate deficiency.
Adverse Reactions
Neuro-B's 500: Thiamine Mononitrate (Vitamin B1): anaphylaxis.
Pyridoxine Hydrochloride (Vitamin B6): long term administration of large doses is associated to the development of severe peripheral neuropathies, sensory neuropathy, paresthesia, somnolence, low serum folic acid levels.
Cyanocobalamin (Vitamin B12): may aggravate Leber's disease or tobacco amblyopia, allergic hypersensitivity, gasping syndrome in premature infants.
Side Effects
Neuro-B's: Vit. B1 + B6 + B12 (Neuro-B's) are completely non-toxic even in high doses and usually, no side effects occur. Sensitivity to Vitamin B1 may sometimes be encountered. Sensitivity may occur if administration is at prolonged at intervals, is irregular, or if the initial dose is low and is suddenly increased: liver dysfunction also seems to predispose to patients to hypersensitivity reaction. Nevertheless, it must be stressed that the hypersensitivity to vitamin B1 is very rare when given orally and is encountered more often when given by intravenous injection.
Drug Interactions
Neuro-B's: Vitamin B6 (pyridoxine hydrochloride) reduces the therapeutic effects of Levodopa.
Neuro-B's 500: Pyridoxine Hydrochloride (Vitamin B6) reduces effects of levodopa (unless a dopa decarboxylase inhibitor is also given) and altretamine; decreases serum concentrations of Phenobarbital; requirement is increased when given concurrently with isoniazid, penicillamine, and oral contraceptives; hydralazine may increase pyridoxine requirements.
Cyanocobalamin (Vitamin B12) absorption from the gastrointestinal tract is reduced in the presence of aminoglycosides, anticonvulsants, biguanides, chloramphenicol, cholestyramine, cimetidine, methyldopa and potassium salts. Inhalation of the anesthetic agent nitrous oxide, anesthetic can produce a functional Vitamin B12 deficiency and colchicines; decreased serum concentrations with concurrent administration of oral contraceptives.
