Indications/Uses
Acute or chronic, peripheral & cerebral vasculo-metabolic disorders w/ signs & symptoms of cerebral insufficiency or dementia eg, impaired memory & conc, dizziness, depressed mood, impaired self-care & sleep-walking cycle, disorientation in time & place, tinnitus, blurred vision, performance insufficiency, headache & fatigue including those due to cerebral thrombosis, embolism & arteriosclerosis in its different manifestations. Signs & symptoms of mental deterioration eg, cognitive, affective, behavioural & somatic disturbances associated w/ cerebral deterioration (chronic cerebrovascular insufficiency &/or senile & pre-senile dementia, Parkinson's disease) eg, decreased vigilance, apathy, interpersonal relationships, asthenia, anorexia. 10 mg: Obliterative arteriopathies of the limbs: Raynaud's syndrome & all syndromes due to altered peripheral flow. Headache in geriatric arteriosclerotic patients. Adjuvant to therapy of arterial HTN. 30 mg: Co-adjuvant in rehabilitative treatment of hemiplegic patients w/ ictus sequelae.
Dosage/Direction for Use
10 mg sugar-coated tab 10 mg tid. 30 mg FC tab 60 mg daily in 2 administrations at regular intervals.
Administration
10 mg sugar-coated tab: Should be taken on an empty stomach: Take on an empty stomach. 30 mg FC tab: May be taken with or without food: Preferably taken before meals.
Contraindications
Hypersensitivity to nicergoline or ergot alkaloids. Recent MI, acute hemorrhage, orthostatic hypotension & severe bradycardia.
Special Precautions
Pulmonary, cardiac, cardiac valvular & retroperitoneal fibrosis. Symptoms of ergotism including nausea, vomiting, diarrhea, abdominal pain & peripheral vasoconstriction. May lower systolic & diastolic BP in normotensive patients & those w/ elevated BP. Patients w/ hyperuricemia or history of gout &/or during concomitant treatment w/ drugs influencing uric acid metabolism & excretion. Concomitant use w/ sympathomimetic α & β agonists. May affect ability to drive & use machines. Pregnancy. Not recommended during lactation.
Adverse Reactions
Drug Interactions
May potentiate effect of antihypertensives; cardiac effect of β-blocking agents. May antagonize vasoconstrictor effect of sympathomimetics. Concomitant use w/ drugs metabolized by CYP2D6. Prolonged bleeding period w/ antiaggregant & anticoagulants (eg, ASA). May increase serum uric acid levels w/ drugs that interfere w/ uric acid metabolism & excretion.
