Cilostazol
Indications/Uses
Management of peripheral vascular disease, often for atherosclerotic or occlusive arterial disease but covers both arterial & venous disorders & may be due to atherosclerosis, vasospasm or thromboembolism.
Dosage/Direction for Use
50 mg or 100 mg bid. Response to treatment may occur in 2-4 wk, but up to 12 wk may be required.
Administration
Should be taken on an empty stomach: Take at least 30 min before or 2 hr after meals.
Contraindications
Hypersensitivity. Patients w/ CHF of any severity; known predisposition to bleeding, history of ventricular arrhythmias, QT interval prolongation, severe renal impairment or moderate to severe hepatic impairment.
Special Precautions
Avoid use or reduce dose in patients taking CYP3A4 or CYP2C19 inhibitors. Pregnancy.
Adverse Reactions
Headache, dizziness, palpitations, diarrhoea; oedema, nausea & vomiting, other cardiac arrhythmias, chest pain, rhinitis, ecchymosis, skin rashes.
Drug Interactions
Concomitant use w/ other drugs that inhibit or are metabolized by CYP3A4 or CYP2C19 may result in changes in plasma conc of cilostazol or the concomitant drug & possibly adverse effects.
