Metformin HCl
Dosage/Direction for Use
Adult Monotherapy & combination w/ other oral antidiabetic agent Starting dose: 500 mg or 850 mg bid-tid. Slowly increase by an increment of 1 tab after 10-15 days depending on blood glucose measurements. Max daily dose: 3 g in 3 divided doses. Combination w/ insulin Starting dose: 500 mg or 850 mg bid-tid, while insulin dosage is adjusted on the basis of blood glucose measurements. Childn ≥10 yr & adolescent Monotherapy & combination w/ insulin Starting dose: 500 mg or 850 mg once daily. Slowly increase by an increment of 1 tab after 10-15 days depending on the basis of blood glucose measurements. Max daily dose: 2 g in 2 or 3 divided doses.
Administration
Should be taken with food: Take w/ or after meals.
Contraindications
Diabetic coma, ketoacidosis, predisposition to lactic acidosis, severe infection or trauma, dehydration, alcohol dependence, deficiencies of vit B12, folic acid & Fe. Should not be given to patients w/ recent MI, dehydration, or any other condition likely to predispose to lactic acidosis. Hepatic or renal impairment. Not advised during pregnancy & lactation.
Special Precautions
Long term administration may cause disturbance of vit B12 absorption.
Adverse Reactions
Anorexia, nausea & diarrhea; metallic taste; wt loss.
Drug Interactions
Increased risk of lactic acidosis & hypoglycemia w/ alcohol. Concomitant use w/ drugs that may impair renal function. Impaired absorption of vit B12.
