Loperamide HCl
Indications/Uses
Non-specific acute & chronic diarrhea related w/ refractory inflammatory bowel disease in patients ≥18 yr; w/ ileostomy.
Dosage/Direction for Use
Acute diarrhea Initially 2 tab, then 1 tab after every defecation. Max: 8 tab daily. Chronic diarrhea Initially 2 tab & continued until hard stools 1-2 times daily which is obtained by dose of 1-6 tab daily. Max: 8 tab daily.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. Patients w/ acute dysentery, acute ulcerative colitis, bacterial enterocolitis due to invasive organisms, pseudomembranous colitis related w/ the use of broad-spectrum antibiotics. Infant <24 mth.
Special Precautions
Discontinue use if there is blood in the stool, fever, or worsening symptoms of flatulence; if early symptoms of abdominal distension appear in AIDS patients; if constipation, abdominal distension, or ileus occurs. Fluid & electrolyte replacement therapy is essential in patients w/ diarrhea. Caution when driving the vehicle or operating the machinery. Closely monitor for signs of CNS toxicity in patients w/ hepatic impairment.
Adverse Reactions
Skin rash, dry mouth, abdominal pain, bloating, nausea, vomiting, flatulence, dyspepsia, constipation, ileus, megacolon, urinary retention, drowsiness, dizziness, abnormal coordination, depressed level of consciousness, hypertonia, decreased level of consciousness, stupor, miosis, fatigue.
Drug Interactions
Concomitant use w/ quinidine, ritonavir, saquinavir, itraconazole, ketoconazole, CYP3A4 inhibitor, P-gp, oral desmopressin.
