Each film-coated caplet contains Sildenafil citrate 100 mg.
50 mg to be taken as needed 1 hour before sexual activity. It may be taken from 4 hour to 0.5 hour before sexual activity. Based on effectiveness and toleration, the dose may be increased to maximum recommended dose of 100 mg or decreased to 25 mg. Maximum dosing frequency is once daily.
Hypersensitivity. Patients who are using organic nitrates, either regularly and/or intermittently.
Men for whom sexual activity is inadvisable because of their underlying cardiovascular status. Patients who have suffered MI, stroke, life-threatening arrhythmia within 6 months. Patients with resting hypotension (BP <90/50) or hypertension (BP 170/110). Cardiac failure or coronary artery disease causing unstable angina. Retinitis pigmentosa, anatomical deformation of the penis, predisposition to priapism. Concomitant administration with ritonavir. Non-arteritic anterior ischaemic optic neuropathy (NAION), a cause of decreased vision or loss of vision. Serious cardiovascular, cerebrovascular and vascular events, including myocardial infarction, sudden cardiac death, ventricular arrhythmia and hypertension. Dose >25 mg should not be taken within 4 hours of taking an alpha-blocker. Discontinue treatment in case of sudden decrease or loss of hearing.
Headache, flushing, dyspepsia, nasal congestion, abnormal vision, diarrhea, dizziness, rash.
CYP3A4/2C9 inhibitors may reduce sildenafil clearance. Increased plasma concentration with cimetidine. Systemic exposure is increased with erythromycin. Reduced clearance with ketoconazole, itraconazole, erythromycin and cimetidine. Cmax and plasma AUC increased with saquinavir, ritonavir.
