Indications/Uses

Erectile dysfunction.

Dosage/Direction for Use

Adult 50 mg as needed, taken 1 hr before sexual activity. May be taken 4 to ½ hr before sexual activity. Dose may be increased to a max of 100 mg or decreased to 25 mg. Max dosing frequency is once daily.

Administration

May be taken with or without food.

Contraindications

Hypersensitivity. Patients either regularly &/or intermittently using organic nitrates.

Special Precautions

Discontinue treatment in case of sudden decrease or loss of hearing. Serious CV, cerebrovascular & vascular events, including MI, sudden cardiac death, ventricular arrhythmia & HTN. Cardiac failure or CAD causing unstable angina. Retinitis pigmentosa, anatomical deformation of penis, predisposition to priapism. Non-arteritic anterior ischemic optic neuropathy (NAION). Patients who have suffered MI, stroke, life-threatening arrhythmia w/in 6 mth; w/ resting hypotension (BP <90/50) or HTN (BP 170/110). Not to take >25 mg dose w/in 4 hr of taking an α-blocker. Concomitant use w/ ritonavir. Men w/ underlying CV status for whom sexual activity is inadvisable.

Adverse Reactions

Headache, flushing, dyspepsia, nasal congestion, abnormal vision, diarrhea, dizziness, rash.

Drug Interactions

Reduced clearance w/ CYP3A4/2C9 inhibitors. Increased plasma conc w/ cimetidine. Increased systemic exposure w/ erythromycin. Reduced clearance w/ ketoconazole, itraconazole, erythromycin & cimetidine. Increased C_max & plasma AUC w/ saquinavir, ritonavir.
