Indications and Dosage
Topical/Cutaneous
Acne vulgaris
Adult: Each g of gel contains clindamycin 1% (10 mg) and benzoyl peroxide 2.5% (25 mg)
Each g of gel contains clindamycin 1% (10 mg) and benzoyl peroxide 3% (30 mg)
Each g of gel contains clindamycin 1% (10 mg) and benzoyl peroxide 5% (50 mg)
Each g of gel contains clindamycin 1.2% (12 mg) and benzoyl peroxide 3.75% (37.5 mg)
For the treatment of mild to moderate cases: Apply a thin film onto the affected area(s) once daily in the evening. Max treatment duration: 12 weeks.

For the treatment of inflammatory cases: Apply a thin film onto the affected area(s) once daily in the evening. Dosage and treatment recommendations may vary among countries or individual products (refer to specific product guidelines).

Contraindications
Hypersensitivity to clindamycin, lincomycin, or benzoyl peroxide. History of regional enteritis, ulcerative colitis, pseudomembranous or antibiotic-associated colitis.
Special Precautions
Atopic patients. Concomitant use with other topical acne treatment. Pregnancy and lactation.
Adverse Reactions
Significant: Anaphylaxis and allergic reactions; skin irritation.
Nervous system disorders: Pareasthesia.
Skin and subcutaneous tissue disorders: Pruritus, burning sensation, dryness, erythema, peeling, worsening of acne.
Potentially Fatal: Pseudomembranous colitis, Clostridioides difficile-associated diarrhoea (CDAD).
Patient Counseling Information
Avoid contact with the eyes, lips, mouth, mucous membranes, or broken skin. This drug may cause an increased sensitivity to sunlight, avoid or minimise prolonged exposure to sunlight and avoid the use of sunlamps; use protective clothing and sunscreen during therapy. May cause discolouration of hair and dyed fabrics.
Drug Interactions
May cause cumulative irritant effect with other topical acne therapy.
Clindamycin: May decrease the therapeutic effect with erythromycin. May enhance the action of neuromuscular blocking agents.
Benzoyl peroxide: May cause yellow/orange discolouration of the skin and facial hair with topical sulfone products (e.g. dapsone, sulfacetamide). May decrease the efficacy of and increase the risk of irritation with tretinoin, isotretinoin, and tazarotene.
Action
Description:
Mechanism of Action: Clindamycin is a lincosamide antibacterial agent. It binds reversibly to the 50S ribosomal subunits preventing the formation of peptide bonds, thereby inhibiting bacterial protein synthesis.
Benzoyl peroxide is an oxidising agent that has keratolytic properties and bactericidal effects. It releases free-radical oxygen which oxidises bacterial proteins in the sebaceous follicles, thus reducing the number of anaerobic bacteria and reducing irritating-type free fatty acids.
Pharmacokinetics:
Absorption: Clindamycin: Minimal absorption through the skin.
Benzoyl peroxide: Absorbed through the skin.
Metabolism: Benzoyl peroxide: Metabolised in the skin to benzoic acid.
Excretion: Benzoyl peroxide: Via urine.
