Indications and Dosage
Oral
Dysmenorrhoea
Adult: For the symptomatic relief of primary dysmenorrhoea: 60-120 mg 1-3 times daily.
Child: ≥12 years Same as adult dose.

Oral
Gastrointestinal spasm
Adult: For the symptomatic relief of spastic functional gastrointestinal disturbances, including diverticular disease of the colon and irritable bowel syndrome: 60-120 mg 1-3 times daily.
Child: ≥12 years Same as adult dose.

Contraindications
Paralytic ileus, intestinal obstruction.
Special Precautions
Patient with nausea or vomiting, anorexia, weight loss, pallor, fatigue, fever, blood from the bowel, severe constipation, abnormal vaginal discharge or bleeding, and dysuria. Patients who have recently travelled abroad. Children. Pregnancy and lactation.
Adverse Reactions
Significant: CNS effects (e.g. dizziness), hepatic effects (e.g. ALT and AST elevations).
Gastrointestinal disorders: Nausea.
Hepatobiliary disorders: Hepatitis.
Immune system disorders: Anaphylaxis, hypersensitivity reaction.
Nervous system disorders: Headache.
Respiratory, thoracic and mediastinal disorders: Dyspnoea, wheezing.
Skin and subcutaneous tissue disorders: Pruritus, skin rash.
Patient Counseling Information
This drug may cause dizziness, if affected, do not drive or operate machinery.
Overdosage
Symptoms: Hypotension, atropine-like toxic effects. Management: Supportive treatment for hypotension. Manage atropine-like symptoms as for atropine poisoning.
Action
Description:
Mechanism of Action: Alverine is an antispasmodic agent. It has a direct action on the smooth muscle of the alimentary tract and uterus, without affecting the heart, blood vessels, or trachea.
Pharmacokinetics:
Absorption: Rapidly absorbed from the gastrointestinal tract. Time to peak plasma concentration: 1-1.5 hours (para hydroxy alverine [PHA]).
Metabolism: Rapidly metabolised into PHA, the primary active metabolite; undergoes further conversion to form 2 secondary metabolites.
Excretion: Mainly via urine (as metabolites). Elimination half-life: 0.8 hours (alverine); 5.7 hours (PHA).
