Indications/Uses
Female infertility in anovulation [including polycystic ovarian syndrome (PCOS) in women unresponsive to treatment w/ clomifene citrate] & controlled ovarian hyperstimulation to induce the development of multiple follicles in medically assisted reproduction programs (eg, in vitro fertilization/embryo transfer, gamete intra-fallopian transfer & intracytoplasmic sperm inj). Spermatogenesis deficiency due to hypogonadotrophic hypogonadism in males.
Dosage/Direction for Use
Individualized dosage. Female Anovulation Starting dose: 50 IU daily maintained for at least 7 days. If there is no ovarian response, gradually increase daily dose until follicle growth &/or plasma oestradiol levels indicate an adequate pharmacodynamic response. Maintain daily dose until pre-ovulatory conditions are reached. Controlled ovarian hyperstimulation in medically assisted reproduction programs Starting dose: 100-225 IU for at least the 1st 4 days. May individually adjust dosage based upon ovarian response. Can be given either alone or in combination w/ a GnRH agonist or antagonist (for premature luteinisation prevention). Higher total Puregon treatment dose may be required to achieve an adequate follicular response, when using a GnRH agonist. Male 450 IU wkly, preferably in 3 divided doses, given concomitantly w/ hCG. Continue treatment for at least 3-4 mth.
Contraindications
Hypersensitivity. Ovarian, breast, uterine, testicular, pituitary or hypothalamic tumors. Primary gonadal failure. Females: Undiagnosed vag bleeding. Ovarian cysts or enlarged ovaries, not related to PCOS. Reproductive organ malformations & uterine fibroid tumors incompatible w/ pregnancy. Pregnancy & lactation.
