Indications and Dosage
Topical/Cutaneous
Corticosteroid-responsive dermatoses
Adult: For relief of inflammatory and pruritic manifestations: As 0.1% cream or ointment: Apply a thin film to the affected area(s) 2-3 times daily, depending on the severity of the condition.
Child: Treatment recommendations may vary among countries and between individual products (refer to specific product guidelines).
Special Precautions
Avoid use of occlusive dressings in the presence of infected or weeping lesions. Children. Pregnancy and lactation.
Adverse Reactions
Significant: HPA axis suppression or hypercortisolism (particularly in younger children or in patients receiving high doses for prolonged periods); allergic contact dermatitis; Kaposi's sarcoma (prolonged use); systemic effects (e.g. Cushing's syndrome, hyperglycaemia, glycosuria).
Skin and subcutaneous tissue disorders: Skin irritation (including burning), itching, dryness, folliculitis, hypertrichosis, acneiform eruptions, hypopigmentation, perioral dermatitis, skin maceration, skin atrophy, striae, miliaria.
Patient Counseling Information
Avoid contact with the eyes. Do not use with occlusive dressings unless directed by the doctor.
Action
Description:
Mechanism of Action: Amcinonide is a highly potent topical corticosteroid that has anti-inflammatory, antipruritic, and vasoconstrictive properties. It may suppress the formation, release, and activity of endogenous inflammatory chemical mediators (e.g. kinins, histamine, liposomal enzymes, prostaglandins) via induction of phospholipase A2 inhibitory proteins (lipocortins) and subsequent inhibition of the release of arachidonic acid.
Pharmacokinetics:
Absorption: Adequately absorbed through intact skin; increased absorption with skin inflammation or use of occlusive dressings.
Metabolism: Metabolised in the liver.
Excretion: Via urine and faeces.
