Human coagulation factor VIII
Indications/Uses
Prophylaxis & treatment of bleeding in congenital &/or acquired factor VIII deficiency in haemophilia A.
Dosage/Direction for Use
Dosage is individualized based on the severity of factor VIII deficiency, location & extent of bleeding & patient's clinical condition. Early haemarthrosis, muscle or oral bleeding 20-40% 12-24 hrly for at least 1 day. More extensive haemarthrosis, muscle bleeding or haematoma 30-60% 12-24 hrly for 3-4 days. Life threatening haemorrhages 60-100% 8-24 hrly. Minor surgery including tooth extraction 30-60% 24 hrly for at least 1 day. Major surgery (pre- & postop) 8-24 hrly. Then, repeat therapy for at least 7 days to maintain 30-60% factor VIII activity.
Contraindications
Pregnancy & lactation.
Special Precautions
Regular monitoring of Ab formation. Consider hepatitis A & B vaccination.
Adverse Reactions
Rarely, hypersensitivity or allergic reactions, headache, hives, drop in BP, lethargy, nausea, restlessness, accelerated pulse, chest tightness, tingling, vomiting, wheezing, anaphylaxis, fever.
